Description: HAP1 (huntingtin-associated protein 1) binds to huntingtin (1). Huntingtin is a protein that contains a polyglutamine region and when the number of glutamine repeats exceeds 35, the gene encodes a version of huntingtin that leads to Huntington’s disease (HD) (2,3). The ability of HAP1 to bind to huntingtin is enhanced by an expanded polyglutamine repeat region (1). HAP1 shows neuronal localization and moves with huntingtin in nerve fibers (4,5). HAP1 is primarily expressed in brain tissue, with greater expression in the olfactory bulb and brain stem (1). Mouse HAP1 is localized to membrane-bound organelles including large endosomes, tubulovesicular structures and budding vesicles in neurons (6). Duo, also designated huntingtin-associated protein interacting protein or HAPIP, binds Huntingtin-associated protein 1 (HAP1) and may have a role in vesicle trafficking and cytoskeletal function.

Description: The homeobox protein, HESX1, which is also known as Rathke’s pouch homeobox, HANF, homeodomain transcription factor, and anterior-restricted homeobox protein is a transcription factor that belongs to the homeodomain family of DNA binding proteins. HESX1 is initially expressed in embryonic stem cells and the primitive forebrain, and is essential for normal development of the eyes and other anterior CNS structures, such as the hypothalamus, the pituitary gland and the olfactory bulbs. The homeobox gene Hesx1 is expressed in the anterior visceral endoderm (AVE), anterior axial mesendoderm (AME), and anterior neural ectoderm (ANE) during early embryogenesis. Mutations in the Hesx1 gene are associated with disorders that are comparable with septo-optic dysplasia (SOD). These disorders are characterized by hypoplasia of the optic nerve, various types of forebrain defects and pituitary hormone deficiencies, including hypothyroidism. Hesx1 also acts as a transcriptional repressor of reporter gene constructs in tissue culture assays.

Description: The Testican family, also designated the BM-40/SPARC/osteonectin family, is composed of highly conserved, extracellular, calcium-binding, sulfate proteoglycans. Expression of Testicans is detected in a variety of tissues, but is most abundant in brain. Family members include Testican-1, Testican-2, Testican-3 and an amino-terminal splice variant of Testican-3, designated N-Tes. Most Testicans inhibit MT-MMPs, thereby inhibiting the activity of pro-MMP-2. Testican-2 is expressed in the central nervous system (CNS), with widespread expression in the olfactory bulb, cerebral cortex, thalamus, hippocampus, cerebellum and medulla, and is also found in lung and testis. Testican-2 is unique in that it actually abolishes the inhibition of MT-MMPs by other testican family members and specifically inactivates N-Tes by binding to its COOH-terminal extracellular calcium-binding domain. Testican-2 halts neurite growth from cerebellar neurons and may be involved in regu-lating the development of the CNS.
