Description: Designed to screen ADAM17 (TACE) inhibitors using a quenched fluorogenic peptide. The ADAM17 Fluorometric (also known as fluorimetric) Drug Discovery Kit is a complete assay system designed to screen ADAM17 (TACE) inhibitors using a quenched fluorogenic peptide: Mca-PLAQAV-Dpa-RSSSR-NH2. Mca fluorescence is quenched by the Dpa group until cleavage by proteases separates the two moieties. The assays are performed in a convenient 96-well microplate format. The kit is useful to screen inhibitors of ADAM17, a potential therapeutic target. The compound GM6001 is also included as a prototypic control inhibitor.

Description: Nerve terminal probes are a series of fluorescent cationic styryl dyes developed to follow synaptic activity. These dyes have a lipophilic tail at one end and a hydrophilic cationically charged head group at the other end. The dyes vary in the number of carbons in the lipophilic tail (m) and the number of double bonds linking the two aromatic rings in the dye (n). AM dyes are similar to FM dyes, but contain an additional formaldehyde-fixable amino group in the cationic head.

The Fluor de Lys® Substrate, which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes the substrate so that, in a second step, treatment with the Fluor de Lys® Developer produces a fluorophore. Histone deacetylases from all known classes have been successfully assayed with the Fluor de Lys® Substrate.
