Description: Cytochrome P450 proteins are heme-thiolate monooxygenases that mediate NADPH-dependent electron transport and function to oxidize a variety of structurally unrelated compounds, including steroids, fatty acids and xenobiotics. Specifically, Cytochrome P450s are responsible for metabolizing arachidonic acid to hydroxyeicosatetraenoic acid (a regulator of blood pressure) and epoxyeicosatrienoic acid (a molecule involved in signaling events). CYP20A1 (cytochrome P450, family 20, subfamily A, polypeptide 1), also known as CYP-M, is a 462 amino acid single-pass membrane protein that belongs to the cytochrome P450 family. CYP20A1 is thought to carry its own oxygen as it lacks a conserved I-helix motif and one amino acid of its conserved heme binding site.

Description: The conversion of arachidonic acid to prostaglandin H2 (PGH2) by cyclooxygenase (COX) enzymes is the first step in the synthesis of prostanoids. Prostaglandin D2 (PGD2) is synthesized from PGH2 by the action of PGD synthase, and its actions in vasodilation and platelet activation are mediated via activation of two identified G-protein coupled receptors (GPCRs): DP1 and DP2 (formerly CRTH2). Both DP1 and DP2 receptors are coupled to Gs alpha subunits, resulting in increased adenylyl cyclase activity and cAMP production upon agonist binding. DP1 receptor is expressed moderately in the ileum and weakly in the lung, stomach, and uterus of mice, and weakly in the small intestine of humans. In rodents, expression of PGD synthase and the DP1 receptor is seen in the leptomeninges, where it is believed they play a role in the induction of sleep. The DP1 receptor may also oppose DP2-mediated activation of basophils, eosinophils, and Th2 cells.

Supplier: ENZO LIFE SCIENCES
Description: Prostaglandin E2 (PGE2) is an extensively studied prostaglandin owing to its predominance in inflammation, cancer, atherosclerosis, autoimmune disease, and sepsis. Oxidation of arachidonic acid by prostaglandin synthases (COX-1 and COX-2) produces prostaglandin H2 (PGH2), which is further metabolized by PGE synthases into its major product, PGE2. PGE2 mediates autocrine and paracrine signaling by binding to G-protein coupled receptors (EP1, EP2, EP3, EP4) on the cell surface, functioning to modulate phospholipase C and adenylate cyclase activity. PGE2 has been of great interest as a therapeutic target, either by modulation of its synthesis by COX inhibitors (NSAIDS) or by modulation of its receptors by downregulation or binding antagonists. PGE2 production in a variety of tissues has been shown to modulate numerous physiological processes including natriuresis in the kidney, smooth muscle elasticity in the vasculature, and the inflammatory response to damaged tissues by monocytes and macrophages.

Catalog Number: (ENZOADI905793100)
Supplier: ENZO LIFE SCIENCES
Description: Leukotrienes are metabolic products of arachidonic acid formed by the action of 5-lipoxygenase (5-LO), with the principal products being leukotrienes C4 (LTC4) and B4 (LTB4). LTB4 is a dihydroxy-leukotriene produced in a variety of cells including eosinophils, neutrophils, platelets, and vascular cells, and is a potent leukocyte chemoattractant known to regulate leukocyte activation, cytokine secretion, inflammatory gene transcription, and IgE synthesis. As such, LTB4 has been implicated in a number of disease pathologies including atherosclerosis, asthma, psoriasis, and inflammatory bowel disease. The many actions of LTB4 are associated with two identified mammalian receptors, BLT1 and BLT2. BLT1 is a high-affinity LTB4 receptor expressed on peripheral leukocytes, while BLT2 is a more ubiquitously expressed low affinity LTB4 receptor whose function is not well understood. The BLT1 receptor is transiently expressed by antigen-primed T-cells in humans, and its expression is known to play a critical role in collagen-induced arthritis in mice.

Description: The complement component proteins, C3, C4 and C5, are potent anaphylatoxins that are released during complement activation. Binding of these proteins to their respective G protein-coupled receptors, C3aR, C1R and C5aR, induces proinflammatory events, such as cellular degranulation, smooth muscle contraction, arachidonic acid metabolism, cytokine release, leukocyte activation and cellular chemotaxis. Complement Factor B, also designated Properdin Factor B or PBF2, is part of the alternate pathway of the complement system and is cleaved by Factor D into two fragments: Ba and Bb. Bb combines with complement Factor 3b to produce the C3 or C5 convertase and plays a role in the differentiation and proliferation of preactivated B lymphocytes, lysis of erythrocytes, stimulation of lymphocyte blastogenesis and rapid spreading of peripheral blood monocytes. Ba is important in inhibiting the proliferation of preactivated B lymphocytes. Adipsin, also designated complement Factor D, is a serine protease that cleaves complement Factor B and may be involved in obesity. Factor H controls the function of the alternative complement pathway. FHR-1 (complement Factor H related protein 1) may play a role in lipid metabolism.
