Description: Angiotensin Converting enzyme is involved in catalyzing the conversion of angiotensin I into a physiologically active peptide angiotensin II. Angiotensin II is a potent vasopressor and aldosterone-stimulating peptide that controls blood pressure and fluid-electrolyte balance. This enzyme plays a key role in the renin-angiotensin system. ACE converts angiotensin I to angiotensin II by release of the terminal His-Leu, this results in an increase of the vasoconstrictor activity of angiotensin. Also able to inactivate bradykinin, a potent vasodilatator. ACE exists in two forms, a 170KD somatic form and a 90KD germinal form. The somatic form is expressed by endothelial cells (especially those of lung capillaries and arterioles), epithelial cells (especially in proximal renal tubules and small intestine), by some neuronal cells and variably by some macrophages and T lymphocytes. The germinal form is expressed by spermatozoa.

Supplier: MP Biomedicals
Description: Pepstatin A is a reversible inhibitor of aspartic proteases, pepsin, renin, cathepsin D, cathepsin G, and other acid proteases. It does not inhibit thiol proteases, neutral proteases or serine proteases.
Catalog Number: (PRSI29-567)
Supplier: ProSci Inc.
Description: Renin catalyzes the first step in the activation pathway of angiotensinogen--a cascade that can result in aldosterone release, vasoconstriction, and increase in blood pressure. Renin, an aspartyl protease, cleaves angiotensinogen to form angiotensin I, which is converted to angiotensin II by angiotensin I converting enzyme, an important regulator of blood pressure and electrolyte balance. Mutations in this gene have been shown to cause familial hyperproreninemia.Renin catalyzes the first step in the activation pathway of angiotensinogen--a cascade that can result in aldosterone release, vasoconstriction, and increase in blood pressure. Renin, an aspartyl protease, cleaves angiotensinogen to form angiotensin I, which is converted to angiotensin II by angiotensin I converting enzyme, an important regulator of blood pressure and electrolyte balance. Transcript variants that encode different protein isoforms and that arise from alternative splicing and the use of alternative promoters have been described, but their full-length nature has not been determined. Mutations in this gene have been shown to cause familial hyperproreninemia.

Description: KLKB1 is a 638 amino acids protein that belongs to the peptidase S1 family and plasma kallikrein subfamily. It contains a peptidase S1 domain and four apple domains. The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. It participates in the surface-dependent activation of blood coagulation, fibrinolysis, kinin generation and inflammation.Plasma prekallikrein deficiency causes a prolonged activated partial thromboplastin time in patients.

Description: BNP (brain natriuretic peptide) belongs to a family of structurally similar peptide hormones, which includes atrial natriuretic peptide (ANP), BNP, C-type natriuretic peptide (CNP) and urodilatin. ANP and BNP act mainly as cardiac hormones, produced primarily by the atrium and ventricle, respectively, while the gene encoding C-type natriuretic peptide is expressed mainly in the brain. BNP circulates in blood as a peptide hormone with natriuretic, vasodilatory and renin inhibitory properties. It is secreted predominantly by the left ventricular myocytes in response to volume expansion and pressure overload. These peptides are characterized by a common 17 amino acid ring structure with a disulfide bond between two cystein residues. This ring structure shows high homology between different natriuretic.

Description: HELSS is a potent, irreversible, mechanism-based inhibitor of Ca2+-independent PLA2 (70% inhibition at 100 nM). It possesses a 1000-fold selectivity for Ca2+-independent PLA2 vs. Ca2+-dependent PLA2 . It has been used to identify the PLA2 involved in the release of arachidonate induced by vasopressin stimulation of A-10 smooth muscle cells (IC50=1 µM). It has also been shown to inhibit the Mg2+-dependent phophatidate phosphohydrolase from P388D1 macrophages (IC50~8 µM).
