Description: Programmed cell death protein 1(PDCD1) is a single-pass type I membrane protein and contains 1 Ig-like V-type domain. PD-1 is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN- gamma by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PDCD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PDCD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. As a cell surface molecule, PDCD1 regulates the adaptive immune response. Engagement of PD-1 by its ligands PD-L1 or PD-L2 transduces a signal that inhibits T-cell proliferation, cytokine production, and cytolytic function.

Description: Alpha-synuclein is a presynaptic neuronal protein that is thought to be involved in the formation of SNARE complexes. Most significantly, aggregated alpha-synuclein is one of the major components found in the Lewy bodies that occur in Parkinson’s disease (PD) and other neurodegenerative disorders (Okochi et al., 2000). Mutation of the alpha-synuclein gene is associated with familial forms of PD (Newmann et al., 2002). Recent evidence suggests that phosphorylation of alpha-synuclein at ser129 plays a role in the formation of inclusions in PD (Smith et al., 2005; Masliah et al., 2005).

Description: CD274, also known as B7-H1 or programmed death ligand 1 (PD-L1), is a 40 kD type I transmembrane protein and a member of the B7 family within the immunoglobulin receptor superfamily. Programmed death-1 ligand-1 (PD-L1, CD274, B7-H1) has been identified as the ligand for the immunoinhibitory receptor programmed death-1(PD1/PDCD1) and has been demonstrated to play a role in the regulation of immune responses and peripheral tolerance. By binding to PD1 on activated T-cells and B-cells, PD-L1 may inhibit ongoing T-cell responses by inducing apoptosis and arresting cell-cycle progression. Accordingly, it leads to growth of immunogenic tumour growth by increasing apoptosis of antigen specific T cells and may contribute to immune evasion by cancers. PD-L1 thus is regarded as promising therapeutic target for human autoimmune disease and malignant cancers.

Description: The TY25 antibody reacts with mouse B7-DC (PD-L2), a 42 kDa glycoprotein of the B7 family of the immunoglobulin superfamily. B7-DC is expressed by macrophages, monocytes, and sub-populations of dendritic cells. B7-DC has similarities with the rest of the B7 family, but it does not bind to CD28/CD152. The receptor for B7-DC is PD-1, which contains an Immunoreceptor Tyrosine-based Inhibitory Motif (ITIM), and is expressed on activated B and T cells. The interaction between B7-DC and its receptor seems to downregulate immune responses. However, B7-DC seems to be a costimulator of T lymphocytes, reacting with a receptor different than PD-1. B7-DC can bind and stimulate dendritic cells. It is also the first B-7 ligand involved in bidirectional communication. The TY25 monoclonal antibody blocks the binding of B7-DC to its receptor (PD-1) and can be used for studying the interaction between the antigen presenting cells and T lymphocytes.

Description: Programmed death ligand 1 (PD-L1, B7-H1 or CD274) is a member of the growing B7 family of immune proteins that provide signals for both stimulating and inhibiting T cell activation. CD274 has been identified as one of two ligands for programmed death 1 (PD-1), a member of the CD28 family of immunoreceptors. CD274 is widely expressed in several organs such as heart, skeletal muscle, placenta and lung, and in lower amounts in thymus, spleen, kidney and liver. CD274 expression is upregulated in a small fraction of activated T and B cells and a much larger fraction of activated monocytes. CD274 expression is also induced in dendritic cells and keratinocytes after IFN-gamma stimulation. CD274 expression is also upregulated in a variety of tumour cell lines. Interaction of CD274 with PD-1 results in inhibition of TCR mediated proliferation and cytokine production, suggesting an inhibitory role in regulating immune responses. The CD274 - PD-1 pathway is involved in the negative regulation of some immune responses and may play an important role in the regulation of peripheral tolerance.
