Description: The MetaCarb™ line of carbohydrate columns provides high resolution columns for applications requiring high performance carbohydrate and organic acid analysis. These columns contain sulfonated polystyrene resins in the Calcium (87C), Hydrogen (87H), and Lead (87P) forms, and provide a wide range of selectivities for carbohydrate and organic analysis. They are widely used in the food and beverage industries for analysis of sweeteners, corn and cane sugars, fruit juices, soft drinks, beer and dairy products.

Description: The Pierce™ NAb™ Spin Kits are convenient for rapid, small-scale affinity purification of antibodies from a variety of sample types. Each pre-filled microcentrifuge spin column of the immobilised protein resin enables quick purification of 100 to 1000 µg of IgG from 25 to 500 µl of serum or other sample. The actual amount of IgG purified varies depending upon the sample type and the specific spin column used.

Supplier: G-Biosciences
Description: The F(ab)₂ Fragmentation kits are designed for the generation and isolation of F(ab)₂ fragments from IgG molecules. The kit utilises Immobilised Pepsin resin. Pepsin is a proteolytic enzyme that is routinely used for the generation of F(ab)₂ fragments from immunoglobulin G (IgG). One or more of the disulfide bonds that join the heavy chains in the hinge region are preserved, so the two Fab regions of the antibody remain joined together, yielding a divalent molecule (containing two antibody binding sites), hence the designation F(ab)₂. The light chains remain intact and attached to the heavy chain, whereas the Fc fragment is digested into small peptides. The Immobilised Pepsin offers the distinct advantage of eliminating enzyme contamination of the F(ab)₂ fragments.

Description: A hands on lab activity to study immunoaffinity chromatography and use a specific antibody to purify antigenic proteins from complex samples. This technique involves performing a chromatography procedure using antigen or antibody immobilised on a chromatographic resin. The solution containing antigen or antibody is passed through the column, which specifically and efficiently captures antibodies (antigen). The captured molecules are eluted from the column as a pure fraction. In this lab activity, students perform immunoaffinity chromatography and learn how this method is utilised in research laboratories.

Description: Phenol is useful for extraction of viral, plasmid and phage DNAs, and to solubilise and dissociate proteins. It is used as a general disinfectant, either in solution or mixed with slaked lime, etc., for toilets, stables, cesspools, floors, drains, etc.; for the manufacture of colorless or light-colored artificial resins, many medical and industrial organic compounds and dyes; as a reagent in chemical analysis. Aqueous solutions are used as topical anesthetic; topical antiseptic; topical antipruritic. Phenol is used in DNA extractions for removing protein.

Supplier: G-Biosciences
Description: Ion exchange chromatography is used to separate charged molecules from complex biological samples. The charged molecules bind to a solid support carrying an opposite charge to the molecule. Proteins contain regions of charged groups on the surface which interact with the ion exchange groups immobilised on the solid support (resin column). Immobilised proteins are eluted by changing either pH or the salt gradient or a combination of both. This lab activity involves preparation of a crude protein extract and running ion exchange chromatography for isolation of proteins.

Supplier: G-Biosciences
Description: Antibody Fab and F(ab)₂ Fragmentation of Mouse IgG1 Kits are designed for the generation and isolation of Fab and F(ab)₂ fragments from mouse IgG₁ molecules. The kit utilises Immobilised Ficin resin. Ficin (or Ficain) (~25000 Da) is a cysteine protease enzyme (EC 3.4.22.3) isolated from fig latex that has the endopeptidase activity to cleave immunoglobulin G molecules in the hinge region. Ficin has an effective range of pH 4 to 9,5 with an optimal pH of 6,5 and cleaves bonds that involve uncharged or aromatic amino acids. Ficin is typically used to cleave mouse IgG₁ as this is difficult to cleave with papain and pepsin. In the presence of 1 to 4 mM or 10 to 20 mM cysteine, ficin generates F(ab)₂ and Fab fragments respectively.

Supplier: VELP SCIENTIFIC
Description: The procedure for determining dietary fibre exposes the sample to a series of enzymatic digestions that simulate the real digestive process which takes place in the human or animal digestive tract, calculating the undigested residue remaining at the end of the analysis. Generally dietary fibre analysis is carried out on foods intended for human consumption whereas raw fibre analysis is carried out on animal feeds or on raw materials of vegetable origin, eg. cereals. The GDE performs enzymatic digestion, a delicate phase where samples are immersed in a thermostatic water bath and stirred. Continuous and constant sample mixing is necessary in order to prevent the sample from overheating. The unit consists of an immersion thermostat, a transparent polycarbonate tank and a VELP 6-place magnetic stirrer to ensure excellent thermoregulation and precision. The use of deionised water is recommended as it eliminates any possible cause of limescale.

Supplier: Thermo Fisher Scientific
Description: GeneJET™ Whole Blood Genomic DNA Purification Mini Kit utilises silica-based membrane technology in the form of a convenient spin column, eliminating the need for expensive resins, toxic phenol-chloroform extractions, or time-consuming alcohol precipitation. The standard procedure takes less than 20 minutes following cell lysis and yields purified DNA greater than 30 kb in size. Isolated DNA can be used directly in PCR, Southern blotting, and enzymatic reactions.

Supplier: Corning
Description: FiltrEX™ filter plates meet the industry standards for plate dimensions. The rigid side walls make the plate ideal for automation and the wide skirt accepts barcodes. Individual filter discs are encapsulated in the plate by a process that ensures 100% integrity of each well. The design of the nozzle prevents sample wicking and cross-contamination. Glass fibre filter plates can be used for a variety of applications, such as plasmid isolation, DNA purification, PCR clean-up or receptor/ligand binding assays. The low-binding hydrophilic PVDF membrane can be used for lysate clarification, protein kinase assays, or bead- or resin-based separation assays.

Supplier: Retsch GmbH
Description: The ZM 200 is a powerful ultra-centrifugal mill for both demanding grinding tasks and efficient routine work. ZM 200 reduces the size of soft to medium-hard and fibrous materials. Ideal for rapid yet gentle preparation of analytical samples of materials such as chemicals, drugs, spices, coal, synthetic resins, plastics, pharmaceutical raw materials and finished products, plant constituents and surfactant determination in washing powders. For grinding larger amounts the use of a paper filter bag or a cyclone with 3 or 5 litre collecting receptacle is recommended. The sample is then cooled by the airstream and discharged more rapidly from the grinding chamber via the passage receptacle.
