Description: TrkA is a member of the neurotrophic tyrosine kinase receptor family. It is a membrane-bound receptor that upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. TrkA is required for high-affinity binding to nerve growth factor (NGF), neurotrophin-3 and neurotrophin-4/5 but not brain-derived neurotrophic factor (BDNF). TrkA leads to cell differentiations and may play a role in specifying sensory neuron subtypes. It has a crucial role in the development and function of the nociceptive reception system as well as establishment of thermal regulation via sweating. SUBUNIT: Exists in a dynamic equilibrium between monomeric (low affinity) and dimeric (high affinity) structures. SUBCELLULAR LOCATION: TrkA is a heavily glycosylated transmembrane protein and contains 13 potential N-linked glycosylation sites. The TrkA-Fc chimera has N-linked and may have O-linked oligosaccharides.

Description: Interleukin 34 (IL-34) was originally identified in humans, by large scale screening of secreted proteins; orthologs have also been found in chimpanzee, murine, rat and chicken. Human IL-34 is synthesized as a 242 amino acid (aa) precursor that contains a 20 aa signal sequence and a 222 aa mature chain. IL-34 is secreted as a homodimer and contains one potential N-glycosylation site.  IL-34 increases proliferation of immune cells known as monocytes; it elicits its activity by binding the Colony Stimulating Factor 1 Receptor (CSF-1R). Expression of IL-34 is most abundant in the spleen and is expressed in many other tissues including the heart, brain, liver, kidney, thymus, intestine and lung. Human IL-34 contains a C-terminal 6x-polyhistidine (6xHis) tag.
