Description: Recognizes 78 kDa moesin protein. Moesin, a member of the talin-4.1 superfamily, is a linking protein of the sub-membranous actin cytoskeleton. It is expressed in variable amounts in cells of different phenotypes such as macrophages, lymphocytes, fibroblastic, endothelial, epithelial, and neuronal cell lines but not in blood cells. The ERM proteins, ezrin, radixin, and moesin are involved in a variety of cellular functions, such as cell adhesion, migration, and the organization of cell surface structures, and are highly homologous, both in protein sequence and in functional activity, with merlin/schwannomin, a neurofibromatosis-2-associated tumor-suppressor protein. Cell lines of epithelial and mesothelial origin contain both moesin and radixin whereas cells of endothelial and lymphoid origin express moesin.

CD8 molecule consists of two chains, termed and chain, which are expressed as a disulphide-linked heterodimer or as an homodimer. CD8 is expressed on T cell subset (cytotoxic/suppressor T cells), thymocytes and NK cells. The majority of CD8 T-cells expresses CD8 as heterodimer. Some subpopulation of CD8 T cells as well as NK cells may express homodimer. CD8 functions as a co-receptor in concert with TCR for binding the MHC class I/peptide complex. The HIV-2 envelope glycoprotein binds CD8 chain (but not chain). The cytoplasmic domain of CD8 associates with p56lck tyrosine kinase.

CD11a complex with the 2 subunit of the integrin family, CD18, to form the cell surface heterodimer, LFA-1 or CD11a /C18 (aLbL). Description: Recognizes a protein of 130-140 kDa, identified as CD22 (also known as BL-CAM). CD22 expression is restricted to normal and neoplastic B cells and is absent from other haemopoietic cell types. In B-cell ontogeny, CD22 is first expressed in the cytoplasm of pro-B and pre-B cells, and on the surface as B cells mature to become IgD . It is not expressed by plasma cells, CD22 is found highly expressed in follicular mantle and marginal zone B-cells, and while germinal center B-cells are relatively weak. CD22 is a member of the immunoglobulin superfamily and serves as an adhesion receptor for sialic acid-bearing ligands expressed on erythrocytes and all leukocyte classes. It also associates with tyrosine kinases and play a role in signal transduction and B-cell activation.

Description: Recognizes an integral membrane glycoprotein of 150 kDa, identified as CD13 (also known as aminopeptidase-N). The antibody recognizes an extracellular epitope. The CD13 antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. CD13 is absent from normal lymphocytes, platelets and erythrocytes. CD13 is also present on fibroblasts; endothelial cells, epithelial cells from renal proximal tubules and intestinal brush border, bone marrow stromal cells, osteoclasts, and cells lining bile duct canaliculi. CD13 is identical to aminopeptidase N (APN), a prominent membrane-bound metalloprotease present on the surface of intestinal brush border and renal tubules. CD13 plays a role in metabolism of biologically active peptides, in phagocytosis, and in bactericidal/tumoricidal activities. It also serves as a receptor for human coronaviruses (HCV). The lineage-restricted pattern of expression of CD13 within the hemopoietic compartment suggests that it may be important in myeloid cell differentiation.

Description: This MAb recognizes a 27 kDa protein, identified as the p27Kip1, a cell cycle regulatory mitotic inhibitor. It is highly specific and shows no cross-reaction with other related mitotic inhibitors. In Western blotting of cell lysates from 7 human breast cancer cell lines (ZR75-1, ZR75-30, MCF-7, MDAMB453, T47D, CAL51, 734B), the antibody labels a single band corresponding to p27Kip1. It functions as a negative regulator of G1 progression and has been proposed to function as a possible mediator of TGF- induced G1 arrest. p27Kip1 is a candidate tumor suppressor gene. Reportedly, low p27 expression has been associated with unfavorable prognosis in renal cell carcinoma, colon carcinoma, breast carcinomas, non-small-cell lung carcinoma, hepatocellular carcinoma, multiple myeloma, and lymph node metastases in papillary carcinoma of the thyroid, as well as a more aggressive phenotype in carcinoma of the cervix.

CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The CD74 molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74 is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, and dendritic cells and by various types of carcinomas. Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17 kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25 kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production.
