Description: T-cell-specific surface glycoprotein CD28, contains an Ig-like V-type domain. CD28 is one of the proteins expressed on T cells that provide co-stimulatory signals, which are required for their activation. It is the receptor for CD80 and CD86. When activated by Toll-like receptor ligands, the CD80 expression is upregulated in antigen presenting cells (APCs). The CD86 expression on antigen presenting cells is constitutive. CD28 is the only B7 receptor constitutively expressed on naive T cells. It involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival.

Description: V-set and immunoglobulin domain-containing protein 4 (VSIG4) is also known as Protein Z39Ig, is a type I transmembrane glycoprotein. VSIG4 is a B7 family-related protein and an Ig superfamily member. VSIG4 contains two Ig-like (immunoglobulin-like) domains. VSIG4 is abundantly expressed in several fetal tissues. In adult tissues, the highest expression of VSIG4 is in lung and placenta. VSIG4 functions as a negative regulator of T cell activation, and may be involved in the maintenance of peripheral T cell tolerance, and is also identified as a potent suppressor of established inflammation. VSIG4 is a phagocytic receptor, strong negative regulator of T-cell proliferation and IL2 production.

Description: Programmed cell death 1 ligand 2 (also known as PD-L2, B7-DC) is a protein that in humans is encoded by the PDCD1LG2 gene. It is mapped to chromosome 9p24.2. PD-L2 is one of two ligands for programmed death-1 (PD-1), a member of the CD28 family of immuno-receptors. Northern blot analysis detect wide expression of PDL2, with highest levels in placenta, heart, pancreas, lung, and liver, and lower levels in spleen, lymph nodes, and thymus. PDL2 negatively regulates T cells and plays an essential role in immune tolerance.

Description: Programmed cell death 1 ligand 2 (PD-L2 or PDCD1 ligand 2) is also known as Butyrophilin B7-DC, CD antigen CD273, which belongs to the immunoglobulin superfamily or BTN/MOG family. The expression of PD-L2 is up-regulated by IFNG/IFN-gamma stimulation in monocytes and induced on dendritic cells grown from peripheral blood mononuclear cells with CSF2 and IL-4. PD-L2 Involved in the costimulatory signal, essential for T-cell proliferation and IFNG production in a PDCD1-independent manner. PD-L2 interaction with PDCD1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production.

Description: Programmed cell death 1 ligand 2 (PD-L2), also known as butyrophilin B7-DC or PDCD1 ligand 2, belongs to the member of B7 family which can regulate the activation and tolerance of T cells. PD-L2 is one ligand for Programmed cell death 1(PD-1), and the other is PD-L1. These two ligands shares 34% aa sequence identity. Mouse PD-L2 gene encodes a 273 amino acids (aa) protein with a putative 19 aa signal peptide, a 201 aa extracellular region , a 21 aa transmembrane domain and a 32 aa cytoplasmic region. The mouse PD-L2 gene is highly expressed in heart, placenta, pancreas, lung and liver while expressed weakly in spleen, lymph nodes and thymus. Besides, the expression of PD-L2 gene can be induced on dendritic cells grown from peripheral blood mononuclear cells under CSF2 and IL4/interleukin-4 treatment, and up-regulated by IFNG/IFN-gamma stimulation in monocytes. PD-L2 usually functions in a PDCD1-independent manner and is involved in regulating costimulatory signal which is essential for T-cell proliferation and IFNG production. Recent studies demonstrate that the expression of PD-L2 on the tumour cells promotes CD8 T cell–mediated rejection of tumour cells, at both the induction and effector phase of antitumour immunity. Moreover, PD-L2 binds to PD-1 cells and enhances T cell killing in a PD-1–independent mechanism.

Description: Butyrophilin-like 2 (BTNL2) is a butyrophilin family member with homology to the B7 costimulatory molecules, polymorphisms of which have been recently associated through genetic analysis; to sporadic inclusion body myositis and sarcoidosis. BTNL2 mRNA is highly expressed in lymphoid tissues as well as in intestine. BTNL2-Ig fusion protein was shown to recognize a putative receptor whose expression on B and T cells was significantly enhanced after activation. It inhibited T cell proliferation and TCR activation of NFAT, NF-kappaB and AP-1 signalling pathways. BTNL2 is the first member of the butyrophilin family that regulates T cell activation, which has implications in immune diseases and immunotherapy.

Description: OX40, also termed CD134 and TNFRSF4, is a T cell co-stimulatory molecule of the TNF receptor superfamily which plays a key role in the survival and homeostasis of effector and memory T cells. OX40 is expressed on CD4+ and CD8+ T cells upon engagement of the TCR by antigen presenting cells along with co-stimulation by CD40-CD40 Ligand and CD28-B7. The interaction between OX40 and OX40 ligand (OX40L) will occur when activated T cells bind to professional antigen-presenting cells (APCs). The T-cell functions, including cytokine production, expansion, and survival, are then enhanced by the OX40 costimulatory signals. OX40 signals are critical for controllling the function and differentiation of Foxp3+ regulatory T cells. OX40-OX40L interaction regulates T-cell tolerance, peripheral T-cell homeostasis, and T-cell-mediated inflammatory diseases.

Description: PDL-1 Antibody: Cell-mediated immune responses are initiated by T lymphocytes that are themselves stimulated by cognate peptides bound to MHC molecules on antigen-presenting cells (APC). T-cell activation is generally self-limited as activated T cells express receptors such as PD-1 (also known as PDCD-1) that mediate inhibitory signals from the APC. PD-1 can bind two different but related ligands, PDL-1 and PDL-2. PDL-1 is a B7-related protein that inhibits cell-mediated immune responses by reducing the secretion of IL-2 and IL-10 from memory T cells. This suggests that PDL-1 may be useful in reducing allogenic CD4+ memory T-cell responses to endothelial cells, thereby reducing the likelihood of host immune responses to allografts. At least two isoforms of PDL-1 are known to exist; this antibody is specific to the larger isoform. PDL-1 antibody has no cross-reactivity to PDL-2.
