Description: CD276, also known as B7-H3, is a member of the B7 superfamily with signature IgV and IgG regions in extracellular domains. It is a type I transmembrane protein and shares 20–27% amino acid identity with other B7 family members. B7-H3 is involved in the activation of T lymphocytes, and regulates murine bone formation. It is also reported that B7-H3 may play an important role in muscle-immune interactions, providing further evidence of the active role of muscle cells in local immunoregulatory processes. B7-H3 is expressed on T-cells, natural killer cells, and antigen presenting cells, as well as some non-immune cells, such as osteoblasts, fibroblasts, fibroblast-like synoviocytes and epithelial cells. High expression of B7-H3 in tumour vasculature also correlates with poor survival in patients, suggesting that it may play a role in tumour cell migration.

Catalog Number: (PRSI90-371)
Supplier: ProSci Inc.
Description: B7-H4 is a B7 family member that negatively regulates T cell immunity by inhibiting T cell proliferation, cytokine production and cell cycle progression. In vitro, B7-H4 inhibits CD4+ and CD8+ T cell proliferation, cytokine production and generation of alloreactive cytotoxic T cells (CTLs). In vivo, blockade of endogenous B7-H4 by specific monoclonal antibody promotes T cell responses. B7-H4 ia an important negative regulator of innate immunity through growth inhibition of neutrophils. B7-H4 is expressed on some tumour cancer cells. The role of B7-H4 in tumour progression may be to transform precancerous cells and then protect them from immunosurveillance.

Description: Human B7-H2 is a member of the growing B7 family of immune costimulatory proteins. Other family members include B7-1, B7-2, B7-H1 (PD-L1), PD-L2 and B7-H3. B7 proteins are members of the immunoglobulin (Ig) superfamily, the extracellular domains contain 2 Ig-like domains and all members have short cytoplasmic domains. B7-H2 has been identified as the ligand for ICOS, a member of the CD28 family of costimulatory receptors. Human B7-H2 is expressed constitutively on resting B cells, dendritic cells and at low levels on monocytes. The B7-H2/ICOS interaction appears to play roles in T cell dependent B cell activation and Th differentiation.

Description: CD80 (B7-1) and CD86 (B7-2) together with their receptors CD28 and CTLA-4 constitute one of the dominant costimulatory pathways that regulate T cell and B cell responses. Although both CTLA-4 and CD28 can bind to the same ligands, CTLA-4 binds to B7-1 and B7-2 with a 20-100 fold higher affinity than CD28 and is involved in the down-regulation of the immune response. B7-1 is expressed on activated B cells, activated T cells and macrophages. B7-2 is constitutively expressed on interdigitating dendritic cells, Langerhans cells, peripheral blood dendritic cells, memory B cells, and germinal center B cells. Additionally, B7-2 is expressed at low levels on monocytes and can be up-regulated through interferon-gamma. B7-1 and B7-2 are both members of the immunoglobulin superfamily. It has been observed that both human and mouse B7-1 and B7-2 can bind to either human or mouse CD28 and CTLA-4, suggesting that there are conserved amino acids which form the B7-1/B7-2/CD28/CTLA-4 critical binding sites.
