Description: beta -Ureidopropionase is a cytoplasmic protein which belongs to the CN hydrolase family of BUP subfamily. beta -Ureidopropionase binds one zinc ion per subunit, catalyzes the last step in the pyrimidine degradation pathway. beta -Ureidopropionase can convert N-carbamyl-beta-aminoisobutyric acid and N-carbamyl-beta-alanine to beta-aminoisobutyric acid and beta-alanine, ammonia and carbon dioxide, respectively. The pyrimidine bases uracil and thymine are degraded via the consecutive action of dihydropyrimidine dehydrogenase (DHPDH), dihydropyrimidinase (DHP) and beta-ureidopropionase (UP) to beta-alanine and beta aminoisobutyric acid, respectively. Defects in beta -Ureidopropionase are the cause of beta -Ureidopropionase deficiency that is characterized by muscular hypotonia, dystonic movements, scoliosis, microcephaly and severe developmental delay.

Description: Voltage-gated K+ channels are important determinants of neuronal membrane excitability. Moreover, differences in K+ channel expression patterns and densities contribute to the variations in action potential waveforms and repetitive firing patterns evident in different neuronal cell types (Maletic-Savatic et al., 1995; Pongs, 1999; Blaine and Ribera, 1998; Burger and Ribera, 1996). The Kv3.1 potassium channel is expressed at high levels in neurons that characteristically fire rapid trains of action potentials (Gan et al., 1999). Particularly high levels of this channel are found in neurons of the auditory brainstem. These neurons appear to participate in neural circuits that determine the intensity and timing of auditory stimuli and use this information to determine the location of sounds in space (von Hehn et al., 2004).

Description: Tryptophan hydroxylase (TPH) catalyzes the 5-hydroxylation of tryptophan, which is the first step in the biosynthesis of indoleamines (serotonin and melatonin) (Martinez et al., 2001). In mammals, serotonin biosynthesis occurs predominantly in neurons which originate in the Raphe nuclei of the brain, and melatonin synthesis takes place within the pineal gland. Description: Alpha-internexin is a Class IV intermediate filament originally discovered as it co-purifies with other neurofilament subunits. Alpha-internexin is related to but distinct from the better known neurofilament triplet proteins, NF-L, NF-M and NF-H, having similar protein sequence motifs and a similar intron organization. It is expressed only in neurons and in large amounts early in neuronal development, but is down-regulated in many neurons as development proceeds. Many classes of mature neurons contain alpha-internexin in addition to NF-L, NF-M and NF-H. In some mature neurons alpha-internexin is the only neurofilament subunit expressed. Antibodies to alpha-internexin are therefore unique probes to study and classify neuronal types and follow their processes in sections and in tissue culture. In addition, recent studies show a marked up-regulation of alpha-internexin during neuronal regeneration. The use of antibodies to this protein in the study of brain tumors has not been examined to date, but is likely to be of interest. Recently Cairns et al. used this antibody to show that alpha-internexin is an abundant component of the inclusions of neurofilament inclusion body disease (NFID), a serious human neurodegenerative disorder. The antibody was also used to confirm the presence of circulating auto-antibodies to alpha-internexin in the sera of some patients with endocrine autoimmunity, as well as in some normal individuals.

Description: Fractalkine(CX3CL1) is a single-pass type I membrane protein and belongs to the intercrine delta family. It consists of an extracellular NH2-terminal domain, a mucin-like stalk, a transmembrane alpha helix, and a short cytoplasmic tail. CX3CL1 exists in two forms: as a membrane-anchored or as a shed 80-95K glycoprotein. Soluble CX3CL1 is generated by limited proteolysis on the cell surface, and a disintegrin and metallopeptidase 10 (ADAM10) and ADAM17/tumour necrosis factor- alpha-converting enzyme (ADAM17/TACE) participate in this shedding. It has been suggested that ADAM10 acts in the constitutive shedding, and ADAM17 acts in response to cell activation. The protein may play a role in regulating leukocyte adhesion and migration processes at the endothelium.
